Baird Parker Agar Base is a culture medium recommended for the detection and enumeration of Staphylococcus, particularly coagulase-positive strains, in food and other materials. The addition of tellurite and egg yolk after sterilization helps differentiate pathogenic Staphylococcus colonies by producing black colonies and clear lipolysis zones due to lecithinase activity. For the coagulase test, one vial of RPF Lyophilized Supplement (DSHB3019) should be added to 90 ml of Baird Parker Agar Base, following the supplement’s instructions.
